Overview

11-[(3-chloro-5-(trifluoromethyl)phenyl)carbamoylamino]undecanoic acid is a synthetic aryl-urea fatty acid (AUFA) derivative investigated as a mitochondrial-targeting anticancer agent. It consists of a 3-chloro-5-(trifluoromethyl)phenyl urea moiety conjugated to an 11-carbon (undecanoic acid) fatty acid chain. The molecule is designed to selectively accumulate in the mitochondria of cancer cells, where it disrupts the mitochondrial membrane potential and induces the generation of reactive oxygen species (ROS), ultimately triggering apoptotic cell death. In preclinical studies, it has demonstrated cytotoxic activity against triple-negative breast cancer (TNBC) cell lines, such as MDA-MB-231. This compound was developed as part of a structure-activity relationship (SAR) study to determine how alkyl chain length affects mitochondrial disruption and cell-killing efficiency, with research indicating that the C11 chain length provides effective mitochondrial targeting, although longer chains like the C13 analogue (7c) may offer enhanced potency.
